There is a direct bus from San José to Manuel Antonio; the ride takes 3 to 5 hours. You can also get to Quepos and take a local bus or taxi.
By car
Route 27 to Orotina, then the Costanera Sur (Route 34) to Quepos. Parking available.
By plane
Local Sansa flights from San José to Quepos (XQP), ~25 minutes.
